Today i wake up earlier before subuh comes. The cold air doesn’t make me cancel my plan to do all my activities this morning. 45 minutes before praying, it’s enough for reread the jurnal that I have downloaded last night and make plan for a new method that I will do today and discuss with my lecturer.

8.00 wib, I leave my room to college, wow it’s a new record for me going to college so earlier in this holiday season.  Its still quiet in the laboratory, I’m the first student who come to lab, there are just laboratory assistant who clean the work place. Maybe I should do the same thing with my work desk, since it always gets wet with the coconut Oil.

08.30 I start my work with a new method on my log book. I haven’t discussed about it with my lecturer but I think I want to try this before I meet him, maybe I can get something interesting result.

First step: I take some ml of buffer solution that I make yesterday and put into IEC column, 1 problem occur. The resin should be remain into the solution for a while time, but the solution doesn’t want to flow into the bottom of column, so its still be at top of the resin. After thinking some alternative way to make it flow, finally the problem can be solved.

Second step: take some ml of elution buffer and put into the column, guess ?? yes, same problem occur, but I have found the solution of this problem.  (Thank GodJ)

Third step: feed the coconut oil into the column. Before I do that step I know that same problem will occur too or maybe worse than before. It’s because oil has a higher viscosity than buffer solution.  ( hmm I should find another way to solve this problem). Then I decided to try the third step into separation funnel, I hope it can work.

14.30 wib, it is time to take out the oil from the funnel, and problem still occur. There is no oil come out from the discharge, maybe it being trap with the resin ( so I can’t collect any sample today) L

What should I do with this???

Maybe I should use compressor to make vacuum condition so the oil will be flow into the discharge, shouldn’t I?

Or maybe u have a better idea that can solve my problem???

What a exhausted day…I arrive at my dorm at 16.00 wib and feel  so languid and weak.. I remember I haven’t eat anything since I wake up in the morning cos I’m fasting today..hehehe…

Tonight, with  full energy I have to find a way to solve my problem and do it tomorrow.
